Recent data from Dr. Ardav\u00edn laboratory have allowed to explain how peritoneal immune cells, free in a fluidic environment in homeostasis, control bacterial infections through the formation of mesothelium-bound dynamic aggregates, formed by macrophages, B1 cells and neutrophils dependent on extravascular coagulation-triggered fibrin polymerization, that provide a physical scaffold allowing the interaction and function of peritoneal immune cells (Immunity, 2021). He trained in general and respiratory medicine at Addenbrookes Hospital Cambridge, Royal Brompton Hospital, and John Radcliffe\/Churchill Hospitals in Oxford, returning to NHLI as Senior lecturer in 1988. Before taking up his present position in 2004, he was Reader in Upper Respiratory Medicine.<\/p>\n<p>He has studied the basic mechanisms of allergic rhinitis and asthma and the influence of treatment. This has involved the development of pharmacodynamic models of allergen challenges in the skin, nose, and lungs and studies of seasonal hay fever. A particular focus has been translational studies of allergen immunotherapy that have elucidated mechanisms of human antigen-specific tolerance and informed novel treatment approaches. He is the principal investigator for several international trials that have resulted in Europe-wide registration of an Alum-based grass pollen vaccine for injection and a sublingual grass allergen tablet, the first allergy vaccine to be registered in the UK for 35 years.<\/p>\n<p>Professor Durham is the current President of the British Society for Allergy and Clinical Immunology and a member of the Immune Tolerance Network Steering Committee of the National Institutes of Health. He is the former Chair of the Research Committee of the World Allergy Organisation and a council member of Collegium Internationale Allergologicum. He has given prestigious lectures including the Dr John Salvaggio and Dr Harold Nelson Lectureships at the American Academy of Allergy, Asthma and Immunology, the John P McGovern Medal from the American College of Allergy, Asthma and Immunology. From the BSACI has been awarded the Jack Pepys Lectureship and William Frankland Medal for outstanding services to clinical allergy. Allergens associated with the saprophytic fungus, <em>Alternaria alternata<\/em> exacerbate airway inflammation in sensitized asthma patients. Exposure of the airway epithelium to <em>Alternaria <\/em>induces oxidative stress, leading to the secretion of ATP, a primary alarmin that stimulates Ca<sup>2+<\/sup> uptake, producing sustained increases in intracellular [Ca<sup>2+<\/sup>]. In the present study, we examined the mechanisms responsible for allergen-evoked DNA fragmentation and release from human airway epithelial cells and determined the role of extracellular DNA (eDNA) in type 2 immunity. Human bronchial epithelial (hBE) cells were used to investigate allergen-evoked DNA mobilization and release using comet assays to measure DNA fragmentation, double-stranded DNA assays to measure the kinetics of eDNA release, and DNA sequencing to determine eDNA composition. The role of eDNA in type 2 immunity was investigated in mice following intranasal exposure to <em>Alternaria<\/em>.<\/p>\n<p><em>Alternaria<\/em> extract rapidly stimulated mitochondrial and nuclear DNA release from hBE cells, whereas mitochondrial DNA, but not nuclear DNA release, was induced by house dust mite extract. <em>Alternaria<\/em>-induced increases in intracellular [Ca<sup>2+<\/sup>] activated the pro-protein convertase enzyme furin, which in turn produced non-canonical activation of caspase-3, ultimately leading to nuclear DNA fragmentation as shown in comet assays. eDNA displayed disproportionally higher amounts of promotor and exon sequences and lower intron and intergenic regions compared to predictions of random DNA fragmentation, indicating that the eDNA release was not a consequence of epithelial cell ETosis. In mice, pretreatment with polyamidoamine dendrimers, which function as DNA scavengers, blocked <em>Alternaria<\/em>-evoked type 2 immune responses. Furthermore, in caspase-3-deficient mice, <em>Alternaria<\/em>-stimulated eDNA release was inhibited. Lastly, co-administration of mouse genomic DNA along with <em>Alternaria<\/em> extract amplified IL-5 and IL-13 secretion into bronchoalveolar lavage fluid, whereas DNA alone produced no response.
