aneurysms remain unsuitable for EVT. Furthermore, clinical outcomes of endovascular intervention for complex lesions, such as large or giant thrombosed aneurysms, are not always satisfactory.<br \/>\nWhile microsurgical intervention for these challenging cases is technically demanding, the integration of bypass surgery and skull base techniques can ensure safer and more definitive (radical) treatment. This study assessed disparities between clinician evaluations and patient-reported outcome measures (PROMs) regarding pain, motor function, and learning difficulties among youth with FNSD. Sixty-two youth with FNSD participated in the study, all of whom reported experiencing pain, motor problems, and\/or learning difficulties. Clinicians also assessed these domains, resulting in a two-by-two categorization matrix: (1) <em>agreement<\/em>: child and clinician report &#8220;problems&#8221;; (2) <em>agreement<\/em>: child and clinician report &#8220;no problems&#8221;; (3) <em>disagreement:<\/em> child reports &#8220;problems&#8221; while the clinician does not; and (4) <em>disagreement:<\/em> clinician reports &#8220;problems&#8221; while the child does not. Prevalence differences in agreement\/disagreement were analyzed. No significant differences in prevalence were observed between evaluators regarding pain (clinician-85%, child-88%), motor (clinician-98%, child-95%), or learning problems (clinician-69%, child-61%). Over 80% of children and clinicians reported pain and motor problems. Instances where both child and clinician reported learning problems (40.3%) exceeded cases where both reported no problems (9.6%) or where only the child reported problems (20.9%). Overall, agreement between pain and motor function assessments was high (&gt;90%), whereas agreement concerning learning difficulties was moderate (49.9%). Disagreement in pain\/motor assessments was minimal (&lt;5%), whereas for learning difficulties, disagreement rates were higher (&gt;20%). In conclusion, a significant concordance exists between PROMs and clinicians&#8217; assessments of pain and motor problems. However, the higher frequency of disagreement regarding learning difficulties emphasizes the importance of incorporating both patient and clinician evaluations in pediatric FNSD treatment.<br \/>\n<strong>Biography:<br \/>\n<\/strong>Dr. Sharon Barak is a senior faculty member and Deputy Head of the Department of Nursing at Ariel University, where she teaches Anatomy and Physiology. She is also a researcher in the Pediatric Rehabilitation Department at Sheba Medical Center, Tel Hashomer. Her expertise lies in clinical exercise physiology and rehabilitation sciences. The AcPb isoform is resultant from alternate splicing of the AcP transcript. Our previous studies using AcPb null (AcPb<sup>-\/-<\/sup>) mice characterized its participation in sleep regulation and emergent neuronal\/glial network properties. Here, we investigated the impact of acute sleep disruption (SD) on brain cancer-related pathways in wild-type (WT) and AcPb<sup>-\/-<\/sup>\u00a0mice, employing RNA sequencing methods. In WT mice, SD increased AcPb mRNA levels, but not AcP mRNA, confirming prior similar work in rats. Transcriptome and pathway enrichment analyses demonstrated significant alterations in cancer, immune, and viral disease-related pathways in WT mice after SD, which were attenuated in AcPb<sup>-\/-<\/sup>\u00a0mice including multiple upregulated Src phosphorylation-signaling-dependent genes associated with cancer progression and metastasis. Our RNAseq findings, were analyzed within the context of The Cancer Genome Atlas Program (TCGA) data base; revealing an upregulation of sleep- and cancer-linked genes (e.g., IL-17B, IL-17RA, LCN2) across various tumors, including brain tumors, compared to normal tissues. Sleep-linked factors, identified through TCGA analyses, significantly impact patient prognosis and survival, particularly in low-grade glioma (LGG) and glioblastoma multiforme (GBM) patients. Overall, our findings suggest that SD promotes a pro-tumor environment through AcPb-modulated pathways.<br \/>\n<strong>Biography:<br \/>\n<\/strong>Dr. Yool Lee is a chronobiology researcher whose work centers on the molecular and cellular mechanisms linking circadian rhythms to physiology and disease. Dr. Lee earned their B.A., M.S., and Ph.D. from Seoul National University in South Korea, where they specialized in circadian clock biology. They completed postdoctoral research at the University of Pennsylvania, discovering key clock-regulating molecules and elucidating their roles in disease processes, particularly cancer. Currently, Dr. Lee serves as an Assistant Professor at Washington State University, where their research investigates how sleep and circadian disruption influence cancer pathogenesis and treatment response, in collaboration with clinicians to develop chronotherapeutic strategies.
