and recurrent traumatic fear memories are central features of stress-related psychiatric disorders, including post-traumatic stress disorder. Memory retrieval can render a consolidated fear memory temporarily labile, after which the memory undergoes reconsolidation. This reconsolidation window provides an opportunity to weaken maladaptive fear memories. However, commonly used behavioral interventions, such as retrieval-extinction procedures, often require repeated exposure to trauma-associated cues and may impose additional stress. Enriched environment exposure represents a mild, non-invasive behavioral intervention that can modulate brain plasticity, but whether it can interfere with fear memory reconsolidation and the underlying circuit mechanisms remains unclear.<br \/>\nIn this study, we investigated whether short-term enriched environment exposure after memory retrieval could disrupt fear memory reconsolidation. Using a conditioned fear memory model, we combined behavioral testing, chemogenetic manipulation, engram cell labeling, viral tracing, immunofluorescence, and single-cell RNA sequencing to examine the behavioral, cellular, circuit-level, and transcriptional mechanisms underlying this effect. Behavioral results showed that short-term enriched environment exposure after fear memory retrieval reduced subsequent fear responses and attenuated both spontaneous recovery and reinstatement, suggesting an impairment of fear memory reconsolidation. Chemogenetic inhibition of dentate gyrus neurons before enriched environment exposure weakened this fear-reducing effect, indicating that dentate gyrus activity is required for enriched environment-mediated reconsolidation interference. Engram cell analyses further showed that enriched environment exposure decreased the number of reactivated dentate gyrus fear engram cells. Moreover, chemogenetic manipulation of dentate gyrus engram cells could mimic or reverse the behavioral effect of enriched environment exposure. Circuit-level experiments revealed that the medial entorhinal cortex and its projection to the dentate gyrus are involved in this process, as inhibition of this pathway impaired enriched environment-induced fear memory reduction. In addition, single-cell transcriptomic analyses suggested that post-retrieval enriched environment exposure induced memory-related transcriptional changes in the dentate gyrus.<br \/>\nTogether, these findings indicate that short-term enriched environment exposure after memory retrieval disrupts fear memory reconsolidation through the medial entorhinal cortex\u2013dentate gyrus circuit and remodeling of dentate gyrus engram cell reactivation. This study provides experimental evidence for a mild, non-pharmacological strategy to weaken maladaptive fear memories and may inform future behavioral interventions for trauma-related psychiatric disorders.<\/p>\n<p><strong>Biography:<br \/>\n<\/strong>Dr. Kai Yuan is an Associate Professor at Peking University Institute of Mental Health and Peking University Sixth Hospital, Beijing, China. His research focuses on the neural mechanisms and intervention strategies of maladaptive emotional and addictive memories. Using behavioral paradigms, neuroimaging, and non-invasive neuromodulation approaches, his work investigates how pathological memories are formed, maintained, retrieved, and modified. His recent studies have examined fear memory reconsolidation, extinction retention, and intervention strategies for reducing the return of fear, with particular attention to the translation of basic neuroscience findings into clinical psychiatry. He has also contributed to research on addiction-related memory and brain mechanisms underlying psychiatric symptoms. By integrating experimental psychology, systems neuroscience, and clinical research, Dr. Yuan\u2019s work seeks to identify circuit-level mechanisms that may be targeted by behavioral or neuromodulatory interventions. Despite this, the interplay between screen exposure, sleep quality, and musculoskeletal outcomes in the Indian paediatric population remains poorly characterised. This study assess these associations from a neurophysiological and behavioural health perspective.\u00a0 Methods: A cross-sectional study was conducted on 252 urban school children aged 11\u201313 years. Screen time was self-reported across recreational and educational domains for in weekdays and weekend. Sleep quality was assessed using the Pittsburgh Sleep Quality Index (PSQI), a validated instrument sensitive to disruptions in sleep onset, maintenance, and subjective sleep quality \u2014 all of which are regulated by circadian and homeostatic neurobiological processes. Muscle Quality Index (MQI) was derived from handgrip dynamometry normalised to BMI. Physical activity (IPAQ-A) and cardiorespiratory fitness (6-min walk test\/VO\u2082max) were also evaluated.\u00a0 Results: Children averaged over 7 hours of daily screen time, substantially exceeding WHO-recommended thresholds. Screen exposure was significantly associated with poorer sleep quality (PSQI positive correlation, p &lt; 0.001), consistent with screen-mediated suppression of melatonin secretion, blue light-induced circadian phase delay, and cortical hyperarousal at bedtime. Boys exhibited significantly worse sleep (PSQI 7.4 vs 5.9, p &lt; 0.01) and higher screen time (7.51 vs 6.8 h, p = 0.007). A significant negative correlation between screen time and MQI (r = \u22120.320, p &lt; 0.01) suggests that neuromotor engagement and musculoskeletal development are compromised by sedentary screen behaviour. In multiple regression analysis, PSQI (\u03b2 = 0.217, p &lt; 0.001), BMI (\u03b2 = 0.208, p = 0.011), and MQI (\u03b2 = \u22120.263, p = 0.019) emerged as independent predictors of screen time, jointly accounting for 23.4% of variance (R\u00b2 = 0.234, p &lt; 0.001).\u00a0 Discussion: The neurobiological mechanisms underpinning these findings are of direct relevance to neuroscience and psychiatry. Screen-induced disruption of the hypothalamic suprachiasmatic nucleus-driven circadian rhythm impairs slow-wave and REM sleep, both critical for synaptic consolidation, emotional regulation, and neurodevelopmental maturation in adolescence. The bidirectional relationship between poor sleep and increased screen use mirrors neuropsychiatric feedback loops observed in paediatric insomnia, attention-deficit\/hyperactivity disorder (ADHD), and anxiety disorders. Moreover, diminished MQI may reflect reduced dopaminergic reward engagement with physical activity, consistent with screen-mediated reward substitution pathways. The absence of a significant correlation with VO\u2082max suggests that cardiorespiratory neural regulation may follow independent pathways, warranting further neuroimaging and polysomnographic investigation.\u00a0 Conclusion: This study demonstrates that excessive screen exposure is associated with disrupted sleep neurophysiology and reduced muscle quality in school-age children. These findings underscore the need for neuro-informed, multidisciplinary interventions targeting screen behaviour, sleep hygiene, and physical activity promotion, with particular urgency in the rapidly urbanising Indian context.<\/p>\n<p><strong>Biography:<br \/>\n<\/strong>Dr. Tejaswini Manogna is a Pediatrician based in India.
