Group B streptococcus (GBS) is a leading cause of invasive neonatal infections and a significant pathogen in immunocompromised adults. Screening of GBS colonization in pregnant women determines the need for antibiotic prophylaxis in that pregnancy. In recent years, there has been rapid expansion in the availability of chromogenic media for the detection of GBS. These culture media contain enzyme substrates linked to indoxyl chromogens, and the target microorganisms are characterized by specific enzyme systems that metabolize the substrate, resulting in release of the chromogen. Subsequently, the indigoid dye formed upon oxidation and dimerization of indoxyl molecules in the presence of oxygen precipitates within the colonies, leading to typical brightly contrasting colors (shown in Fig.b).
Therefore, the amplitude of the contrasting is positively to concentration of GBS. However, the traditional visual identity usually needs at least 72 h to determine whether the GBS colonize, thus, a rapid highly sensitive detection of GBS is urgent. Here, we set up a POCT (shown in Fig.a) with specific spectral absorption (λ2) of chromogenic culture media to replace the traditional visual identity of GBS, the measurable absorbed amount of light is indirectly proportional to the concentration of GBS (shown in Fig.c), which greatly improved the sensitivity of GBS detection, and decreased the time to identify it.

The aim is to navigate the complexities of a diverse and faith-driven landscape.
Cameroon, with its religious diversity and a significant Christian population (69.2%), highlights the need for tailored sexual health education. The CBCHS operates across 41 healthcare facilities in the country, serving various Christian denominations. The author’s extensive CBCHS involvement equips them to offer guidance to fellow advocates and educators in sexual reproductive health.
Respecting Christian values, such as abstinence, purity, and sanctity of marriage, is fundamental. This approach creates an empathetic and inclusive environment, fostering healthier, more informed, and compassionate Christian communities where individuals make faith-aligned sexual health decisions.
Religious leaders’ support is pivotal, as they hold significant influence within their congregations. They serve as spiritual guides and moral authorities, endorsing initiatives involving sensitive topics, contributing to a more compassionate and understanding approach within the faith context.
Balancing cherished values with real-world circumstances is key. Comprehensive sexual health education, while upholding abstinence, addresses situations where abstinence may not be possible. This pragmatic approach aligns with responsible stewardship, guiding individuals to make informed and responsible sexual health choices.
Open and respectful dialogue is central. Encouraging individuals to voice their questions, express concerns, and engage in discussions empowers them to make faith-framed, informed decisions. Objective: To compare sperm quality and quantity, hormonal profiles and spontaneous pregnancy rates before and after administering a 3 months course of N-Acetylcholine (NAC)
Material and Methods: This prospective clinical trial was conducted at the Infertility Unit of the Obstetrics and Gynecology Department at Bhumibol Adulyadej Hospital, Thailand. The study period was from June to September, 2023. Subjects were idiopathic infertile males aged between 20 and 50 years old. All subjects received 600 mg of NAC orally per day. Semen analysis (SA) and male hormonal profiles (MHP; testosterone, LH, FSH and prolactin) were performed before and three months after NAC administration. Demographic characteristics of both couples were collected. Improvement of SA, MHP, and spontaneous pregnancy were also collected.
Results: The 92 participants were recruited. The mean age of male and female participants was 35.1 and 33.8 years old, respectively. The average duration of infertility was 3.6 years. Mean body mass index of subjects was 26.4 kg/m2. Half (50/92) and one-quarter (22/92) of participants were classified as obese and overweight, respectively. One-seventh (12/92) of participants had underlying diseases, namely hypertension, dyslipidemia, and diabetes mellitus. There was a statistically significant increase in semen volume, concentration, and all attributes of motility. Semen volume, concentration, normozoospermia, sperm motility and MHP significantly increased after three months of NAC administration. No changes in sperm morphology and prolactin level. One-third (27/92) of each participant’s spouse conceived spontaneously.
Conclusion: NAC potentially enhances male hormonal profiles, sperm quality and quantity with an impressive spontaneous pregnancy rate.

Background: Although the Positive Discipline Program is used extensively internationally, there are very few evidence-based studies on it. This study aimed to measure the effect of a Positive Discipline Program applied to parents with preschool children on parents’ attitudes, assessment of parenting attitudes of couples, parents’ communication with their children.
Method: A randomized-controlled study was conducted with intervention, active control, and non-contact control groups and pre-, post, and follow-up tests. The study sample consisted of 72 parents with children aged between 3 and 6 years. The intervention group participated in an eight-session online Positive Discipline Program, the active control group participated in an eight-session online free interaction program, and the non-contact control group received no intervention.
Results: Parents in the intervention group demonstrated an increase in cooperation with their children after the training and during the follow-up period. In the intervention group, parents’ oppressive and authoritarian attitudes decreased after the training and during the follow-up period; spouses’ democratic attitudes increased after the training and their overprotective attitudes decreased in the follow-up period. Parent–child communication and unhindered listening increased in the intervention and active control groups. The problem-solving skills of the intervention group increased after the training and during the follow-up period, and sensitivity increased in the follow-up period.
Conclusion: As one of the few experimental studies implementing the Positive Discipline Program, this study revealed the strengths of the program as well as culture-specific aspects that need to be improved. Further, this program was effective in improving parents’ relationships with their children, attitudes, and problem-solving skills, and it provided evidence for the program’s positive contributions to raising a child. These results prove that the Positive Discipline Program is a sustainable, feasible and effective program.
Implications for Practice: The Positive Discipline Program can guide parents in solving the developmental and school-related problems of preschool children as well as school and adolescent children.

Objective: To evaluate Retinopathy of Prematurity (ROP) rate and risk factors in a large cohort of preterm newborns.
Methods: Single center retrospective study. All preterm inborn hospitalized at the Neonatal Intensive Care Unit of the Policlinico of Catania from January 1, 2009 till December 31, 2018, were included.ROP stage and location, treatments required, maternal and infant risk factors were evaluated.
Results: Medical records of 898 preterms were retrospectively examined (mean gestational age 32.9 ± 2.3 weeks). Of them 149 (16.6 %) developed bilateral ROP (92 stage 1, 44 stage 2 and 13 stage 3); 66 (7.3 %) received bilateral laser treatment. Six eyes of three patients affected by zone I ROP 1, with plus persistence 15 days after an optimal laser treatment, also received intravitreal ranibizumab injection. Risk factors for ROP development were gestational age (GA) (p < 0.001), birthweight (p < 0.001), assisted ventilation duration (p < 0.001), multiple birth (p = 0.003), erythropoietin (EPO) administration (p = 0.005) and persistence of tunica vasculosa lentis. The decision-tree analysis showed gestational age as the most significant predictive factor (P < 0.001); secondary predictive factors were EPO administration (p = 0.001) in newborns 29–31 weeks GA and birthweight lower than 2090 g (p < 0.001) in 32–34 weeks GA; in this latter group patent ductus arteriosus (PDA) was a tertiary predictive factor (p = 0.043).
Conclusions: In our study ROP incidence was 16,6 %; 7.3 % of the patients required laser treatment. Besides well-known factors, such as GA and birthweight, other factors like duration of assisted ventilation, EPO, multiple births, PDA, tunica vasculosa lentis persistence should be considered to tailor ophthalmic evaluation and follow-up.

Depression affects approximately 10% to 15% of pregnant and postpartum women and carries significant risks for both mother and baby. There are currently no FDA- approved antidepressants for perinatal depression. Many mothers are reluctant to take medications while pregnant and nursing. Therefore, non-pharmacologic neuromodulation therapies such as transcranial magnetic stimulation (TMS) have gained interest. Approved by the Food and Drug Administration in 2008, TMS is a noninvasive, safe, neuromodulation therapy that uses focally targeted magnetic pulses to modulate neuronal activity in specific brain areas. TMS avoids the risks of seizure, anesthesia and cognitive impairment observed with electroconvulsive therapy (ECT), generates an electric field far below the safety limit recommended in pregnant patients, and avoids risks of maternal medication side effects and fetal medication exposure. Although there are no current guidelines for the use of TMS for perinatal depression, research supports the use of TMS as a safe and effective treatment option during pregnancy and postpartum. Due to the high prevalence and significant negative impact of perinatal depression and to the strong sentiment in pregnant and nursing mothers to avoid medication, there is an urgent need for more research, especially large randomized controlled double-blind trials using standardized TMS protocols, to assess the long-term safety, for both women and children exposed in utero, and efficacy of TMS for perinatal depression.

Objective: To assess the quality of antenatal care and its covariates among postpartum women in Mozambique.
Design: Cross-sectional study.
Setting: The study was undertaken either at the maternity hospital or at home, in Mozambique, in 2019.
Participants: A total of393 women aged 18 to 49 years were interviewed immediately after giving birth.
Measurements: The quality of antenatal care was analysed based on the guidelines of the Ministry of Health of Mozambique: antenatal appointment in the first 16 weeks of gestation, 4 or more antenatal appointments, at least one laboratory test, at least 4 or more clinical-obstetric procedures, 3 doses of the prophylactic intermittent treatment for malaria, 2 doses of the tetanus vaccine, and counselling related to general health care. Women who reported all of these situations were classified as having had quality antenatal care.
Findings: All women reported at least one antenatal care visit, but only 13.0% were classified as having had quality antenatal care. Women who underwent antenatal care in previous pregnancies (aOR=4,28; 95%CI=1,45-12,62) and whose current pregnancy was planned (aOR=2,51; 95%CI=1,11-5,68) were more likely to have quality antenatal care than those who had never had access to antenatal care in previous pregnancies and those who currently had an unplanned pregnancy. It was observed that the criteria recommended by the Ministry of Health were not followed during antenatal care, especially regarding the gestational age at the beginning of antenatal care, the minimum number of consultations, performing blood tests (complete blood count, blood glucose, blood typing and Rh factor) and guidance on signs of risk in pregnancy and childbirth.
Key conclusions: In general, the criteria recommended for quality antenatal care are not incorporated into clinical practice in Mozambique.
Implications for practice: While quality antenatal care is essential to reduce maternal morbidity and mortality, many sub-Saharan countries still have low-quality antenatal care services, which is one of the reasons why these countries have the highest maternal mortality rates worldwide.

Breast cancer, the most common tumor in women in Mali and worldwide has been linked to several risk factors, one of which includes genetic factors such as the PIN3 16-bp duplication polymorphism of TP53. The aim of our study was to evaluate the role of the PIN3 16-bp duplication polymorphism in the susceptibility to breast cancer in the Malian population. Using AS-PCR, we analyzed the PIN3 16-bp duplication polymorphism in blood samples of 60 Malian breast cancer and 60 healthy Malian women (Age-matched study). In this study, no correlation was observed between the PIN3 16-bp duplication polymorphism and clinic-pathological characteristics in the breast cancer patients except histological types (P= 0.04). The genotypic frequency distribution was 45.0% A1A1, 41.7% A1A2, 13.3% A2A2 in cases and 65.0% A1A1, 26.7% A1A2, 8.3% A2A2 in controls. Allele frequencies in cases and controls were 65.8% A1, 34.2% A2 and 78.3% A1, 21.7% A2. respectively. The A1A2 heterozygous genotype was associated with an increased risk of breast cancer with (OR = 2.25, 95% CI = 1.01-5.01 and p = 0.04). According genetic models, a significant association of PIN3 16-bp duplication polymorphism with breast cancer risk was observed in dominant (A1A2+A2A2 vs. A1A : OR = 2.26, CI 95% = 1.08-4.73; P = 0.02) and additive (A2 vs. A1: OR =1.87, CI 95% = 1.05-3.33; P = 0.03) models, but not in the recessive model (A2A2 vs. A1A1+A1A2: 1.69 = 0.52-5.50; P = 0.38). The case-control study showed that PIN3 16-bp duplication polymorphism of TP53 may be a significant risk factor for breast cancer in Malian women.

After lung cancer, breast cancer is the most prevalent cause of mortality all over the world. MTX is a folic acid antagonist that blocks the intracellular enzymatic process that promotes cell proliferation, DNA synthesis, and RNA synthesis to inhibit the cytosolic enzyme Dihydrofolate reductase (DHFR) and alter the catalytic conversion of Dihydrofolate to tetrahydrofolate. Due to negative side effects such drug resistance, the necessity for large doses, non-specific targeting, limited permeability, and short plasma half-life, the utility of MTX for cancer therapy has been constrained. To combat this, by encapsulating pharmaceuticals to NPs, nanotechnology can offer superior cancer therapeutics, perhaps resolving the issue of side effects and drug efflux. In the present investigation, the effect of encapsulated MTX on the apoptosis of breast cancer cells (MCF-7 cell line) was checked using real-time PCR. Moreover, studies on the complex’s physicochemical composition were conducted. 75% of the complex was successfully encapsulated, and the complex exhibited appropriate controlled release performance. When methylthiazol tetrazolium (MTT) was used to measure cell cytotoxicity, it was discovered that magnetic alginate beads-MTX had a stronger anticancer impact than free MTX at lower dosages. Apoptotic markers Bcl2 associated X gene (Bax), B-cell lymphoma 2 (Bcl-2), and neuroinflammatory marker tumor necrosis factor- alpha (TNF-α) genes expression level on the treated cells were assessed using real- time polymerase chain reaction (PCR). The results showed that in MCF-7 cell line, the Bax expression was significantly increased whereas Bcl-2 and TNF levels were decreased. These results indicated that encapsulating MTX in alginate beads might successfully reduce the drug’s dose while having decreased level of negative side effects. Due to their biodegradable and biocompatible nature, magnetic alginate beads coated with glutaraldehyde cross-linked chitosan are a suitable MTX delivery system. Stable magnetic alginate beads react to a magnetic field by acting locally in a cancer cell line. Overall, it can be concluded that usage of alginate beads loaded with MTX can be a novel treatment strategy in the future of cancer therapy and reduce the side effects of chemotherapeutic agents.
